Vitrification of Mouse Oocytes by EM Grid and Cryoloop

Abstract
Objectives: The aim of this study was to compare the survival and developmental rates of mouse oocytes underwent vitrification according to different pre-equilibration period and carriers. Methods: Mouse oocytes obtained from superovulated mice were treated for 2.5 min or 10 min pre-equilibration with 1.5 M ethylene glycol. Oocytes were then cryopreservated by vitrification using EM grid or cryoloop with 5.5 M ethylene glycol plus 1.0 M sucrose. Recovery and survival rates of oocytes after thawing and developmental rates to hatching blastocyst stage after in vitro fertilization were compared between these groups. Results: Investigating the optimal pre-equilibration period for the vitrification by two different carriers, 10 min for pre-equilibration period using EM grid and cryoloop vitrification have been shown higher levels than those of 2.5 min for oocyte recovery, survival, fertilization, and hatching rates. The optimal vitrification carriers by two different equipments, EM grid and cryoloop, for oocyte vitrification were examined. The cryoloop carriers for oocyte vitrification were significantly higher rates, such as recovery (90.3% vs 84.2%), survival (82.8% vs 64.1%) and fertilization (57.8% vs 45.4%) during 2.5 min for pre-equilibration period compared with the EM Grid. Like 2.5 min for pre-equilibration period, 10 min for pre-equilibration period using cryoloop vitrification were significantly higher rates, such as recovery (99.1% vs 87.5%), survival rates (89.5% vs 71.4%) compared with the EM grid vitrification. Conclusion: On growth and development of mouse oocytes, vitrification with cryoloop resulted in better outcome than that with EM grid. In vitrification with EM grid and cryoloop, 10 min of pre-equilibration period resulted in better rates of oocyte recovery, survival, fertilization, and hatching than those of 2.5 min of pre-equilibration period.
